The University require's a single-photon camera array detector capable of spatially-resolved detection of individual visible-wavelength photon events, with temporal precision suitable for TCSPC fluorescence lifetime imaging microscopy. The project will involve microscope imaging of cell-membrane-localised FLIM-FRET tension sensors across a field of view of multiple cells within the beating heart of transparent zebrafish larvae. Analysis of changes in the full time-of-arrival fluorescence lifetime histograms will enable us to determine the lifetime of the fluorescence decay, and infer from this the physical properties of the cells under observation.
